The major difference between Benadryl and ZzzQuil is that two different brands manufacture them. Both Benadryl and ZzzQuil contain Diphenhydramine as the active ingredient. 

Diphenhydramine qualifies as an anti-allergy medication, which also helps to induce sleep. It doesn’t matter who the manufacturer is, the active ingredient of the medication works in the exact same way. 

However, Benadryl and ZzzQuil may differ in dosage strengths and the other ingredients added in their formulations like ZzzQuil contain alcohol which enhances the sedation caused by Diphenhydramine, hence it is named that way. 

Benadryl, on the other hand, does not contain alcohol. Rest assured that they work the same way and you can’t expect a significant difference in the therapeutic outcomes of these two meds. Make sure you use either one of them and not use both of them together. 

Benadryl should be avoided with any other formulation that either contains Diphenhydramine or any other antihistamine- as two antihistamines should not be taken together. NyQuil, a commonly used OTC cold medication, is often paired with Benadryl accidentally. 

Formulations like NyQuil and Unisom contain another sedative antihistamine called Doxylamine, and these meds can increase the risk of certain side effects when taken together with Benadryl. This is why OTC formulations should be taken carefully.

Do Benadryl and ZzzQuil have the same side effect profile?

Yes, Benadryl and ZzzQuil do have the same side effect profile because the active ingredient responsible for causing a pharmacological effect is the same. Different dosage strengths, however, can cause more or less intense side effects. 

Commonly reported side effects associated with the use of Diphenhydramine include:

  • Drowsiness

  • Dry mouth, nose, or throat

  • Chest tightness or wheezing
